Bacteriophages are increasingly used as natural preservatives in food and cosmetics applications. For a cost-effective production of larger amounts of bacteriophages, a scalable and preferably platformed production process is desirable.  

Bacteriophage production: Use of precipitation as a primary recovery step after cell lysis

Currently, many bacteriophage production processes make use of precipitation as a primary recovery step after cell lysis. This step is needed to reduce viscosity of the cell broth, which is primarily caused by a high DNA concentration. Bacteriophages greatly differ in size and outer protein structure. Depending on the phage, precipitation may result in a significant loss of phage number and activity.  

Maximize intact and active phages while minimizing cost & complexity with NuCLEANase® 

Enzymatic digestion of DNA can be used as an alternative to precipitation for viscosity reduction of the cell broth. Enzymes such as NuCLEANase®, can be added just before or during the lysis step. DNA released by the cells will immediately be digested, which reduces product loss by aggregate building. The enzyme is highly specific to its substrate (DNA and RNA) without any proteolytic activity. This makes enzymes a promising purification method to maximize intact, active phages while minimizing cost and complexity.
